Description
Polyoxidonium is a synthetic immunomodulator, proved to have antioxidant activity. Polyoxidonium displays immunomodulatory activity, increases the resistance of organism both to local and general infections. Together with immunomodulatory properties, Polyoxidonium has a pronounced detoxic and antioxidative activities that are determined by its structural and high-molecular nature. The application experience has demonstrated the considerable clinical effectiveness in therapy of chronic, recurrent, indolent infectious-inflammatory disorders of skin, bronchopulmonary apparatus, gastrointestinal and urogenital tracts, as well as in the treatment of severe bacterial and viral infections.

Therapeutic actions:
The effect of polyoxidonium, a synthetic immunomodulator, which is a water soluble high-molecular compound, an N-oxidized polyethylene-piperazine derivative, on the functional activity of human peripheral blood phagocytic cells was studied in vitro. It was established that a one-hour incubation of leukocytes with polyoxidonium increased the ability of leukocytes to kill the ingested Staphylococcus aureus in a dose-dependent manner. This increase is observed with leukocytes obtained both from healthy persons and from patients with chronic granulomatous disease. The study of spontaneous and stimulated chemiluminescence of phagocytes showed a significant decrease in the quantity of chemiluminescent impulses in extracellular space in the presence of polyoxidonium both in luminol- and lucigenin-dependent chemiluminescence. Polyoxidonium proved to have antioxidant activity at all doses tested (100, 250, and 500 μg/ml). The analysis of intracellular hydrogen peroxide (H2O2) level by means of dichlorofluorescein, a fluorescent indicator, showed that incubation with polyoxidonium resulted in a higher luminescence intensity of dichlorofluorescein, thus indicating an increase in the intracellular H2O2 level. This increase is not so considerable as it is in case with phorbol myristate acetate stimulation. However with polyoxidonium used at a dose of 500 μg/ml, the difference with the control is significant both for neutrophils and monocytes. Polyoxidonium can be used as adjuvant in combined treatment of acute and chronic infections of any etiology, in the treatment of CGD and secondary immunodeficiencies simultaneously with basic drugs.
What is it used for?:
(Indications:)
Polyoxidonium is indicated for children and adults in the complex therapy of the immune deficit states in the form of the acute and chronic recurrent infection and inflammatory process of any etiology, which cannot be treated by standard methods of therapy. Polyoxidonium is widely used in the complex therapy in pulmonology, phthisiology, otorhinolaryngology, urinology, gynecology, surgery etc. The medication is used in rheumatology (rheumatoid arthritis); in oncology (in the process of the chemical and radiation therapy of the tumours, for correction of the immune suppression influence of the tumours and specific therapy), for reduction of the nephritic- hepatotoxic action of the chemical medication, for activation of the regeneration processes (fractures, burns, trophic ulcers).
As the monotherapy Polyoxidonium is used:
• For prophylaxis of the post-operational infection complications;
• For correction of the secondary immunodeficiency, resulting from ageing or influence of the unfavorable factors.
• For prophylaxis of ARD (intranasal).
Contraindications and cautions:
Personal intolerance. Pregnancy, lactation (no clinical experience on application). Use with cautions when acute renal insufficiency is registered.
Side effects:
Pain at the site of injection in case of intramuscular application.
Interactions:
Polyoxidonium is compatible with antibiotics, anti-viral, anti-fungal and anti-histamine medications, bronchial spasmolytic, glucocorticoids, cytostatics.
